India made 205 for 5 off their 20 overs, thanks to Captain Rohit Sharma who slammed 92, Suryakumar Yadav 31, Shivam Dube 28 and Hardik Pandya 27.
Mitchell Starc and Marcus Stoinis bagged two wickets each for Australia.
In reply, Australia made 181 for 7 off 20 overs.
Travis Head made 76, captain Mitchell Marsh 37 and Glenn Maxwell 20.
Arshdeep Singh finished with 3 for 37.
India finish top of Super 8s Group 1 with a perfect record and will face defending champions England in the semi-finals in Guyana on Thursday.
Australia will go out if Afghanistan beat Bangladesh in the final group game tonight, or Bangladesh win and significantly boost their net run-rate, with South Africa waiting in the last four.
